| Contexte | Tyrosine-protein phosphatase non-receptor type substrate 1 (CD172a/SIRPA) is a ~54 kDa protein. Immunoglobulin-like cell surface receptor for CD47. Acts as docking protein and induces translocation of PTPN6, PTPN11 and other binding partners from the cytosol to the plasma membrane. Supports adhesion of cerebellar neurons, neurite outgrowth and glial cell attachment. May play a key role in intracellular signaling during synaptogenesis and in synaptic function. Involved in the negative regulation of receptor tyrosine kinase-coupled cellular responses induced by cell adhesion, growth factors or insulin. 1. Yao, M. et al. (2014) Journal of the American Society of Nephrology : JASN 25, 1171-86. PMID: 24511121 |

Detects CD172a/SIRPA in indirect ELISAs.

Flow-cytometry using anti-human CD172a antibody.U937 cells were stained with an irrelevant antibody (Blue Histogram) or an anti-human CD172a antibody monoclonal antibody (Catalog # HX892107 ,Green Histogram) at a concentration of 5 µg/ml for 30 mins at RT. After washing, bound antibody was detected using a FITC conjugated goat anti-mouse antibody (Catalog # MF690414) and cells analysed on a NovoCyte Flow Cytometer.

Flow-cytometry using anti-human CD172a antibody.Human peripheral blood monocytes were stained with an irrelevant antibody (Blue Histogram) or an anti-human CD172a antibody monoclonal antibody (Catalog # HX892107 ,Green Histogram) at a concentration of 5 µg/ml for 30 mins at RT. After washing, bound antibody was detected using a FITC conjugated goat anti-mouse antibody (Catalog # MF690414) and cells analysed on a NovoCyte Flow Cytometer.

Flow-cytometry using anti-human CD172a antibody. U937 cells were stained with an irrelevant antibody (Blue Histogram) or an anti-human CD172a monoclonal antibody (Catalog: HX892107, Yellow Histogram) at a concentration of 5 µg/ml for 30 mins at RT. After washing, bound antibody was detected using a a Goat Anti-Mouse IgG H&L Polyclonal Antibody, FITC (abinScience: MF690414) and cells analysed on a NovoCyte Flow Cytometer.
