Fondo

Xaa-Pro aminopeptidase 3 (XPNPEP3) is a ~57 kDa protein. Catalyzes the removal of a penultimate prolyl residue from the N-termini of peptides, such as Leu-Pro-Ala. Also shows low activity towards peptides with Ala or Ser at the P1 position.

Immunofluorescence

A549 cells were fixed with 4% paraformaldehyde for 20 minutes, followed by blocking with 5% goat serum for 1 hour. The cells were then incubated with a primary antibody targeting XPNPEP3 (HV814014) at a concentration of 41.5 ug/ml overnight at 4°C. Afterward, the cells were incubated with a secondary antibody, Goat Anti-Rabbit IgG (Alexa Fluor-488) for 50 minutes at room temperature. The localization of XPNPEP3 was visualized (shown in green), and nuclear DNA was stained with DAPI (shown in blue).
Western blot

Western blot analysis was performed using anti-XPNPEP3 polyclonal antibody at 1μg/ml on various samples.

Lane 2: HepG2 cell lysate
Lane 3: PC3 cell lysate
Lane 4: Mouse kidney lysate
