Oropouche virus (OROV) is an enveloped, tri-segmented, negative-sense RNA peribunyavirus of the family Peribunyaviridae that causes Oropouche fever — the second most common arboviral febrile illness in Latin America after dengue. OROV is transmitted primarily by the biting midge Culicoides paraensis and has caused major epidemics in Brazil, Peru, and Panama, with an estimated 500,000+ cases since 1955. In 2024, OROV re-emerged with unprecedented geographic expansion across Brazil and cases reported in travelers returning to Europe and the United States, raising global surveillance concern. The M segment encodes the glycoprotein precursor (GPC) cleaved into Gn (G1) and Gc (G2) — Gn mediates receptor attachment and is the primary target for neutralizing antibodies, while Gc drives low-pH-triggered membrane fusion. Nucleoprotein (NP, S segment) is the standard diagnostic antigen.

NP-based ELISA is currently the most accessible diagnostic approach for OROV, given the limited availability of commercial RT-PCR kits. Why is OROV suddenly getting attention in 2024?
OROV underwent unprecedented geographic expansion in 2024, spreading beyond its traditional Amazonian endemic zone to major urban centers in Brazil and producing travel-associated cases in Europe and North America. This expansion, combined with the first documented cases of vertical transmission (congenital OROV) and potential neurological complications, has elevated OROV as a global surveillance priority. There are currently no approved vaccines or antivirals for OROV.

Which reagents are best for building an OROV diagnostic ELISA?
For IgM/IgG serology: use NP protein N-His (VK008012) as the coating antigen — NP is the most immunogenic OROV protein and elicits early IgM responses. For antigen-capture ELISA: pair the anti-NP pAb (VK008014) as capture with a labeled detection antibody, or use Gn C-Fc (VK008021) for Gn-based detection.

Do your OROV nanobodies cross-react with other bunyaviruses?
OROV Gn shares limited sequence identity with other peribunyaviruses (Bunyamwera, Schmallenberg). Cross-reactivity of our anti-Gn nanobodies with related viruses has not been established.
