Lumpy Skin Disease Virus (LSDV) Research Reagents — Recombinant Proteins

Lumpy skin disease virus (LSDV) is a large, enveloped, double-stranded DNA capripoxvirus (~151 kb genome) that causes lumpy skin disease (LSD) in cattle — an OIE/WOAH-notifiable transboundary disease characterized by fever, generalized skin nodules, lymphadenopathy, and reduced milk and meat production. Historically restricted to sub-Saharan Africa, LSDV has undergone dramatic geographic expansion since 2012 — spreading through the Middle East, Turkey, the Balkans, Russia, and into Southeast and East Asia (China 2019, India 2019, Indonesia 2022), becoming one of the most rapidly spreading cattle diseases globally. LSDV is transmitted by blood-feeding arthropods (stable flies, mosquitoes, ticks). Like all poxviruses, LSDV produces two infectious forms: intracellular mature virus (IMV) and extracellular enveloped virus (EEV), each with distinct surface antigens. LSDV encodes ~156 predicted ORFs, including structural proteins (P32/LSDV074, IMV envelope LSDV060, EEV glycoprotein LSDV122), host range factors (LSDV141), immune evasion proteins (GPCR LSDV011), and the RPO30 RNA polymerase subunit used in DIVA diagnostics.

Research Use OnlyNot for diagnostic or therapeutic use.

abinScience offers a unique LSDV recombinant protein panel covering seven key viral proteins — spanning both IMV and EEV surface antigens, the immunodominant P32 antigen, RPO30 (DIVA marker), fusion protein, and the viral GPCR — all in His-tagged formats. This is one of the most complete LSDV protein panels commercially available. Manufactured by AtaGenix Laboratories (ISO 9001 & ISO 13485). Key Research Targets

P32 / LSDV074 — Immunodominant Antigen
P32 (also known as the major envelope protein homolog of VACV H3L) is the most immunodominant LSDV antigen and the primary target for serological ELISA-based LSD diagnostics. Anti-P32 antibodies appear early in infection and are highly abundant in convalescent sera, making P32-based ELISA the most widely used serological assay for LSD surveillance. Available: P32 recombinant protein N-His (VK817012).

IMV Envelope Protein / LSDV060
LSDV060 is the IMV (intracellular mature virus) surface protein — homolog of VACV D8L. IMV antigens are targets for virus-neutralizing antibodies that block primary infection. Available in two formats: C-His (VK756011) and N-His (VK756012).

EEV Glycoprotein / LSDV122 & Host Range Protein / LSDV141
• LSDV122 (EEV glycoprotein, homolog of VACV B5R): EEV surface protein that protects extracellular virions from complement — targets for antibodies that block virus dissemination. Available: C-His (VK607011) and N-His (VK607012)
• LSDV141 (EEV host range protein): determines host range and is involved in EEV formation. Available: C-His (VK610011) and N-His (VK610012)

RPO30 / LSDV035 — DIVA Diagnostic Marker
RPO30 is a 30 kDa RNA polymerase subunit that is the basis for RPO30 PCR-based DIVA assays — distinguishing wild-type LSDV from Neethling-based homologous vaccines (which have RPO30 deletions/mutations). Recombinant RPO30 protein can serve as a positive control for RPO30-based diagnostic development. Available: RPO30 recombinant protein N-His (VK792012).

Fusion Protein / LSDV117 & GPCR / LSDV011
• LSDV117 (fusion protein): involved in cell-to-cell spread — recombinant protein N-His (VK684012)
• LSDV011 (G-protein-coupled chemokine-like receptor): viral GPCR that modulates host immune signaling — a unique immune evasion tool found in capripoxviruses. Available: recombinant protein N-His (VK811012)

Frequently Asked Questions

Which protein is best for LSD serological ELISA?
P32/LSDV074 (VK817012) is the standard choice — P32 is the most immunodominant LSDV antigen with the highest sensitivity and specificity in published ELISA validation studies. Use as coating antigen for indirect ELISA to detect anti-LSDV IgG in cattle sera.

How do I implement DIVA diagnostics for LSDV?
DIVA (Differentiating Infected from Vaccinated Animals) for LSDV uses RPO30-based PCR — Neethling homologous vaccine strains have characteristic RPO30 gene deletions that distinguish them from wild-type LSDV. Our RPO30 protein (VK792012) serves as a positive control for assay validation. For serological DIVA, combining P32 ELISA (detects both infected and vaccinated) with EEV-specific antigens (LSDV122/LSDV141) may provide differentiation — contact us for DIVA assay design support.

Do LSDV reagents cross-react with sheeppox and goatpox viruses?
LSDV, sheeppox virus (SPPV), and goatpox virus (GTPV) are all capripoxviruses sharing >96% nucleotide identity. P32, LSDV060, and LSDV122 homologs are highly conserved across all three viruses — cross-reactivity is expected and can be advantageous for pan-capripoxvirus diagnostics. Species-specific differentiation requires PCR-based approaches targeting variable regions (RPO30, GPCR).
