| 배경 | Programmed cell death 1 ligand 1 (CD274/PD-L1) is a ~33 kDa protein. Plays a critical role in induction and maintenance of immune tolerance to self (1,2,3,4). As a ligand for the inhibitory receptor PDCD1/PD-1, modulates the activation threshold of T-cells and limits T-cell effector response (1,2,3,5). Through a yet unknown activating receptor, may costimulate T-cell subsets that predominantly produce interleukin-10 (IL10) (6). Can also act as a transcription coactivator: in response to hypoxia, translocates into the nucleus via its interaction with phosphorylated STAT3 and promotes transcription of GSDMC, leading to pyroptosis (7) CD274 is the therapeutic target of atezolizumab (Tecentriq) and durvalumab (Imfinzi). Untransfected cells (blue Histogram) and Transfected cells (Yellow Histogram) were stained with an anti-human CD274 monoclonal antibody (Catalog: HV974076) at a concentration of 5 µg/ml for 30 mins at RT. After washing, bound antibody was detected using a Goat Anti-Human IgG H&L Polyclonal Antibody, FITC (abinScience: HF690414) and cells analysed on a NovoCyte Flow Cytometer.
