Bundibugyo virus (BDBV) is a filamentous, enveloped, negative-sense single-stranded RNA filovirus first identified during a 2007 hemorrhagic fever outbreak in Bundibugyo District, Uganda, with a ~25% case fatality rate. BDBV is one of six recognized species in the genus Orthoebolavirus, phylogenetically distinct from the more well-known Zaire ebolavirus (ZEBOV) and Sudan ebolavirus (SEBOV). A second BDBV outbreak occurred in the Democratic Republic of Congo in 2012. Like all ebolaviruses, BDBV encodes seven structural proteins: GP (the sole surface glycoprotein and neutralization target), NP (nucleoprotein), VP40 (matrix, drives budding), VP35 (polymerase cofactor, IFN antagonist), VP30 (transcription activator), VP24 (secondary matrix, IFN signaling inhibitor), and L (polymerase). BDBV-specific reagents are critical because ZEBOV-targeted therapeutics (Inmazeb, Ebanga) may have reduced efficacy against BDBV due to GP sequence divergence.

Envelope Glycoprotein (GP) — Neutralization & Vaccine Target
GP is the sole surface protein mediating viral entry via NPC1 in endosomes. BDBV GP shares ~65% amino acid identity with ZEBOV GP — sufficient for partial cross-reactivity of some broadly neutralizing antibodies but too divergent for reliable protection by ZEBOV-specific therapeutics. Available:

Nucleoprotein (NP) — Diagnostic Antigen
NP is the most abundant viral protein in infected cells and the primary target for filovirus diagnostic assays. BDBV NP can be used to develop BDBV-specific diagnostics and to assess cross-reactivity of pan-ebolavirus NP assays. Available: recombinant NP protein N-His (VK059012), anti-NP recombinant antibody SAA2994 (VK059013), and anti-NP polyclonal antibody (VK059014).
VP40 (Matrix Protein)
VP40 drives virion budding and determines the characteristic filamentous morphology of ebolaviruses. VP40 is also the basis for virus-like particle (VLP) production systems. Available: recombinant VP40 protein N-His (VK058012) and anti-VP40 polyclonal antibody (VK058014).
VP30 & VP24 — Transcription & Immune Evasion
VP30 is a zinc-binding transcription activator unique to filoviruses. VP24 inhibits IFN signaling by blocking STAT1 nuclear import. Both are drug target candidates. Available:

Do Inmazeb (ZEBOV-specific) antibodies work against BDBV?
Inmazeb (atoltivimab + maftivimab + odesivimab) was developed against ZEBOV GP and has limited cross-neutralization of BDBV due to ~35% GP sequence divergence. This is precisely why BDBV-specific and pan-ebolavirus reagents are needed. Our broadly neutralizing antibodies BDBV223 and CA45 neutralize BDBV, ZEBOV, and SUDV — critical tools for developing pan-ebolavirus therapeutics.
