Lumpy skin disease virus (LSDV) is a large, enveloped, double-stranded DNA capripoxvirus (~151 kb genome) that causes lumpy skin disease (LSD) in cattle — an OIE/WOAH-notifiable transboundary disease characterized by fever, generalized skin nodules, lymphadenopathy, and reduced milk and meat production. Historically restricted to sub-Saharan Africa, LSDV has undergone dramatic geographic expansion since 2012 — spreading through the Middle East, Turkey, the Balkans, Russia, and into Southeast and East Asia (China 2019, India 2019, Indonesia 2022), becoming one of the most rapidly spreading cattle diseases globally. LSDV is transmitted by blood-feeding arthropods (stable flies, mosquitoes, ticks). Like all poxviruses, LSDV produces two infectious forms: intracellular mature virus (IMV) and extracellular enveloped virus (EEV), each with distinct surface antigens. LSDV encodes ~156 predicted ORFs, including structural proteins (P32/LSDV074, IMV envelope LSDV060, EEV glycoprotein LSDV122), host range factors (LSDV141), immune evasion proteins (GPCR LSDV011), and the RPO30 RNA polymerase subunit used in DIVA diagnostics.

P32 (also known as the major envelope protein homolog of VACV H3L) is the most immunodominant LSDV antigen and the primary target for serological ELISA-based LSD diagnostics. Anti-P32 antibodies appear early in infection and are highly abundant in convalescent sera, making P32-based ELISA the most widely used serological assay for LSD surveillance.
