Rift Valley fever virus (RVFV) is an enveloped, tri-segmented, negative/ambisense RNA phlebovirus of the family Phenuiviridae that causes Rift Valley fever (RVF) — a zoonotic disease primarily affecting ruminants (sheep, cattle, goats) with devastating economic impact (abortion storms, neonatal mortality), and causing hemorrhagic fever, encephalitis, and retinitis in humans. RVFV is transmitted by Aedes and Culex mosquitoes, with periodic explosive outbreaks across sub-Saharan Africa, Egypt, and the Arabian Peninsula driven by heavy rainfall events. RVFV is a WHO R&D Blueprint priority pathogen with significant bioterrorism concern (CDC Category A). The M segment encodes the glycoprotein precursor cleaved into Gn and Gc. Gn mediates receptor binding and is the primary target for neutralizing antibodies; Gc drives low-pH-triggered class II membrane fusion. The S segment encodes nucleoprotein (NP, diagnostic antigen) and NSs (IFN antagonist, major virulence factor).

Which reagents are best for RVFV diagnostic ELISA development?
For IgM/IgG serology: use NP protein (VK101032) as ELISA coating antigen — NP elicits early and robust antibody responses and is conserved across all RVFV lineages. For antigen-capture or Gn-based neutralization ELISA: pair anti-Gn mAb SAA2775 (VK101013) as capture with anti-Gn pAb (VK101014) as detection.
Why does RVFV have both Gn and Gc glycoproteins?
Like all bunyaviruses, RVFV uses a two-glycoprotein entry system: Gn handles receptor attachment while Gc drives membrane fusion. This is analogous to CCHFV (Gn + Gc) and hantaviruses (Gn + Gc), but distinct from coronaviruses (single S protein) or paramyxoviruses (H/G + F). Can I work with these reagents at BSL-2?
Yes. All RVFV products are recombinant proteins and antibodies — no live or inactivated virus. Standard BSL-1/BSL-2 conditions are sufficient. Live RVFV requires BSL-3+ (BSL-3 Agriculture in the US, BSL-3 in most other countries).
