Crimean-Congo Hemorrhagic Fever Virus (CCHFV) Research Reagents — Antibodies & Recombinant Proteins

Crimean-Congo hemorrhagic fever virus (CCHFV) is an enveloped, tri-segmented, negative-sense RNA nairovirus (~19.2 kb total genome) transmitted by Hyalomma ticks and the causative agent of Crimean-Congo hemorrhagic fever (CCHF) — a severe disease with 10–40% case fatality rate and the most widespread tick-borne viral hemorrhagic fever globally (endemic across Africa, Asia, the Middle East, and southeastern Europe). CCHFV is classified as a WHO R&D Blueprint priority pathogen and a CDC Category C bioterrorism agent. The M segment encodes a glycoprotein precursor (GPC) that is proteolytically processed into Gn, Gc, GP38 (secreted glycoprotein), and mucin-like domain. Gc is the primary target for neutralizing antibodies; GP38 is an emerging diagnostic and therapeutic target; NP (nucleoprotein, S segment) is the standard diagnostic antigen; and the L segment encodes the RNA-dependent RNA polymerase (RdRp).

Glycoprotein Gc — Primary Neutralization Target
Gc is the class II fusion glycoprotein responsible for low-pH-triggered membrane fusion in endosomes. Anti-Gc antibodies are the most potent CCHFV neutralizers and the focus of therapeutic antibody development. Available:

Glycoprotein Gn — Receptor-Binding Subunit
Gn mediates initial attachment to host cell receptors (LDLR family) and partners with Gc for viral entry. Available: recombinant Gn proteins in two formats — C-Strep (VK608021) and C-Fc (VK608031) — plus full-length GP ectodomain N-His (VK608012) and anti-GP polyclonal antibody (VK608014).

GP38 — Secreted Glycoprotein / Diagnostic Target
GP38 is a unique secreted glycoprotein cleaved from the GPC precursor. GP38 is shed from infected cells and detectable in patient serum, making it a promising biomarker for early CCHF diagnosis — potentially complementing NP-based assays. Available: GP38 C-Fc (VK608011), GP38 N-His (VK608042), and anti-GP38 polyclonal antibody (VK608044).

Nucleoprotein (NP) — Primary Diagnostic Antigen
NP is the most abundant protein in CCHFV-infected cells and the standard target for diagnostic ELISA (IgM/IgG serology) and antigen-capture assays. NP is highly conserved across CCHFV genotypes, enabling broad geographic coverage in diagnostics. Available: recombinant NP protein N-His (VK608022) and anti-NP polyclonal antibody (VK608024).

RdRp (RNA Polymerase, L Protein)
The L protein is the RNA-dependent RNA polymerase and a target for broad-spectrum antiviral development (favipiravir, ribavirin). Available: recombinant antibody SAA2808 (VK608053) and anti-RdRp nanobody SAA3032 (VK608063) — enabling RdRp detection and functional studies for antiviral drug development.

Frequently Asked Questions

Which antibodies are best for CCHFV neutralization studies?
Anti-Gc antibodies provide the most potent CCHFV neutralization. We recommend ADI-36121 (VK608033) and ADI-37801 (VK608043) — both derived from human survivor B cells and characterized in published neutralization studies. Combining two clones targeting different Gc epitopes can enhance breadth across CCHFV genotypes.

What is GP38 and why is it important?
GP38 is a secreted glycoprotein unique to nairoviruses — it is cleaved from the GPC precursor and released from infected cells into the bloodstream. Serum GP38 levels have been proposed as an early diagnostic biomarker for CCHF, potentially detectable before seroconversion. Our GP38 C-Fc (VK608011) can be used to develop GP38-capture ELISAs, and the anti-GP38 pAb enables detection in patient sera and cell culture supernatants.

Can I work with these reagents at BSL-2?
Yes. All abinScience CCHFV products are recombinant proteins and antibodies — no live or inactivated virus material. Standard BSL-1/BSL-2 laboratory conditions are sufficient. Live CCHFV requires BSL-4 containment.
