Border disease virus (BDV) is a small, enveloped, positive-sense single-stranded RNA pestivirus of the family Flaviviridae (~12.3 kb genome) that causes border disease (BD) in sheep and goats — characterized by congenital defects (hairy shaker lambs), barren ewes, abortion, and persistently infected (PI) animals that shed virus lifelong. BDV is closely related to bovine viral diarrhea virus (BVDV) and classical swine fever virus (CSFV), with frequent interspecies transmission. The virion encodes three structural proteins — envelope glycoproteins E1 and E2 (forming a heterodimer), and the secreted ribonuclease Erns (E0) — plus non-structural proteins (NS2-3, NS4A/B, NS5A/B). E2 is the immunodominant neutralization target and the primary vaccine antigen. Erns is a unique secreted glycoprotein with RNase activity that suppresses innate immunity by degrading extracellular dsRNA, making it a distinctive diagnostic marker and potential antiviral target.

Erns is a unique pestivirus protein with no homolog in other virus families — a secreted glycoprotein possessing intrinsic RNase T2 activity that degrades extracellular dsRNA to suppress type I/III interferon induction. Erns is detectable in serum of PI animals and serves as a diagnostic antigen for identifying persistent BDV infection. Frequently Asked Questions

Do BDV reagents cross-react with BVDV and CSFV?
BDV, BVDV, and CSFV are closely related pestiviruses with E2 amino acid identity of ~60–70%. Cross-reactivity varies by antibody epitope — conserved epitopes may cross-react, while hypervariable regions are species-specific. What is the difference between Erns and E2 for BDV diagnostics?
E2-based ELISA detects anti-E2 antibodies (seroconversion marker, post-vaccination or infection). Erns-based assays detect the Erns protein itself in serum — Erns is actively secreted by infected cells and is detectable in PI animals that may be seronegative. Using both targets together (E2 serology + Erns antigen detection) provides the most comprehensive BDV diagnostic coverage.

Which E2 protein format is best for serology ELISA development?
E2 C-Fc (VK750041) is recommended for oriented capture on Protein A-coated plates — the Fc tag ensures consistent antigen orientation and maximizes antibody binding sites exposure. For direct coating, E2 C-His (VK054011) is a simpler option with reliable adsorption.
