Porcine reproductive and respiratory syndrome virus (PRRSV) is a small, enveloped, positive-sense single-stranded RNA arterivirus (~15.4 kb genome) that causes PRRS — the most economically devastating infectious disease of the global swine industry, with annual losses estimated at $664 million in the United States alone. PRRSV exists as two genetically distinct species: PRRSV-1 (European/Betaarterivirus suid 1, formerly Lelystad virus) and PRRSV-2 (North American/Betaarterivirus suid 2, formerly VR-2332). The virion encodes seven structural proteins: the major envelope glycoprotein GP5 (neutralization target, forms GP5/M heterodimer), minor glycoproteins GP2, GP3, GP4 (receptor binding via CD163/sialoadhesin), nucleocapsid N (diagnostic antigen), and membrane protein M. Non-structural proteins NSP4 (3CL protease), NSP7, and NSP9 (RdRp) are key targets for antiviral drug development and replication monitoring.

Structural Proteins — Envelope Glycoproteins & Nucleocapsid

Glycoprotein 5 (GP5) — Major Neutralization Target
GP5 is the major envelope glycoprotein and the primary target for neutralizing antibodies. GP5 forms a heterodimer with M protein and contains the immunodominant neutralizing epitope (ectodomain B epitope). GP5 sequence hypervariability drives immune evasion and complicates vaccine design. Available: recombinant GP5 protein N-GST&C-His (VK016012) and anti-GP5 polyclonal antibody (VK016014).

Minor Glycoproteins GP2, GP3, GP4 — Receptor Binding Complex
GP2, GP3, and GP4 form a heterotrimer that mediates receptor binding (CD163, sialoadhesin/CD169). GP4 is the primary receptor-binding subunit and an increasingly important vaccine target. Available:

Nucleocapsid Protein (N/NP) — Diagnostic Antigen
NP is the most abundant viral protein and the standard target for PRRSV ELISA diagnostics (IDEXX PRRS X3, indirect ELISA). NP is highly conserved between PRRSV-1 and PRRSV-2, enabling broad diagnostic coverage. Available: recombinant NP protein N-His (VK752012), recombinant anti-NP antibodies Nb1 (VK752013) and SAA2530 (VK752023), and anti-NP polyclonal antibody (VK752014).

Non-Structural Proteins — Replication & Drug Targets

NSP4 / 3CL Protease
NSP4 is the main viral protease (chymotrypsin-like 3CL protease) responsible for polyprotein processing. NSP4 is a primary target for antiviral drug development — analogous to SARS-CoV-2 Mpro targeted by nirmatrelvir. NSP7 — Immunomodulatory Protein

Our NP protein and anti-NP antibodies are broadly reactive across both PRRSV species due to high NP sequence conservation. Glycoprotein reagents (GP2–GP5) are derived from PRRSV-2 sequences and may have reduced cross-reactivity with PRRSV-1 strains. Which proteins are best for PRRSV DIVA diagnostics?
DIVA (Differentiating Infected from Vaccinated Animals) strategies typically use NP-based ELISA for infection detection (NP antibodies appear early and persist) alongside GP5 or GP2/GP3/GP4-based assays for strain-specific characterization. Our GP2/GP3/GP4 proteins in C-Fc format are ideal for oriented ELISA coating in differential serological assays.

Can the NSP4/3CL protease be used for antiviral drug screening?
