When to choose anti-idiotype vs antigen-capture?

Anti-idiotype offers highest drug specificity for mAbs; antigen-capture is useful when anti-Id is unavailable or to assess target-bound (active) drug states.

What’s the difference between free and total drug?

Free drug quantifies unbound drug; total drug includes bound (to target/ADA) and unbound after acid dissociation per protocol.

ELISA or ECL?

ELISA is cost-effective and widely accessible; ECL provides wider dynamic range and lower background for high-concentration samples.

How to evaluate LLOQ/ULOQ suitability?

Ensure calibration curve covers expected Cmin–Cmax. Lot-specific LLOQ/ULOQ must meet accuracy (80–120%) and precision (CV ≤15–20%) criteria.
