Selection Guide

  1. Mechanism: Choose CD3×TAA for T-cell redirection vs. dual-blockade or receptor co-targeting for pathway modulation.
  2. Format: IgG-like for extended half-life/handling; tandem scFv for compact TCE designs; scFv-Fc/DART-like for balance of size and stability.
  3. Fc Design: Use effector-silent Fc for CD3 engagers to minimize off-target FcγR effects; enable Fc if ADCC/ADCP is part of your hypothesis.
  4. Affinity & Arm Balance: Verify KD/EC50 for both arms; avoid excessive CD3 affinity that may increase background activation.

FAQs

IgG-like vs. tandem scFv—which to choose?

IgG-like offers extended half-life and stability; tandem scFv is compact and ideal for TCEs. For better process stability, consider scFv-Fc or DART-like formats.

What is the optimal CD3 affinity?

Moderate CD3 affinity is typically preferred to achieve effective T-cell activation while minimizing background. Use EC50 and cytokine release data from cell models to guide selection.

Do these have Fc effector function?

Both Fc-silent and effector-enhanced versions are available. Fc-silent is standard for CD3 TCEs; effector-enabled can be selected for dual-blockade or ADCC/ADCP studies.
