The coagulation cascade is a tightly regulated network of serine proteases, cofactors, and inhibitors that maintains hemostatic balance. Dysregulation leads to thrombotic disorders (venous thromboembolism, stroke, DIC) or bleeding conditions (hemophilia, von Willebrand disease). The interplay between coagulation, fibrinolysis, and endothelial biology is also central to cancer-associated thrombosis, sepsis-induced coagulopathy, and cardiovascular disease progression. Characterizing these pathways requires well-validated antibodies, recombinant coagulation factors, and functional assay reagents.

Coagulation Cascade

Tissue Factor (TF / CD142 / F3) — The primary initiator of the extrinsic coagulation pathway. TF binds Factor VIIa to form the extrinsic tenase complex, triggering the coagulation cascade. Aberrant TF expression on tumor cells, activated monocytes, and endothelium links coagulation to cancer-associated thrombosis, sepsis-induced DIC, and atherosclerotic plaque rupture.

von Willebrand Factor (vWF) — Large multimeric glycoprotein essential for platelet adhesion to injured endothelium and stabilization of circulating Factor VIII. vWF deficiency or dysfunction causes von Willebrand disease, the most common inherited bleeding disorder. Anti-vWF antibodies support multimer analysis, ELISA quantification, and functional platelet adhesion assays.

Fibrinogen (FGA / FGB / FGG) — A 340 kDa hexameric glycoprotein and the central substrate of the coagulation cascade. Thrombin cleaves fibrinogen to form fibrin monomers that polymerize into the clot scaffold. Fibrinogen is also a major acute-phase reactant, with elevated levels independently predicting cardiovascular risk.

Fibrinolysis & Regulation

D-Dimer — A fibrin degradation product generated when cross-linked fibrin is cleaved by plasmin. Elevated D-Dimer is the first-line exclusion biomarker for venous thromboembolism (DVT/PE) and is used to monitor DIC severity and thrombolytic therapy response. Anti-D-Dimer antibodies enable rapid immunoturbidimetric and lateral flow assay development.

PAI-1 (SERPINE1) — The primary physiological inhibitor of tissue plasminogen activator (tPA) and urokinase (uPA). Elevated PAI-1 suppresses fibrinolysis, promoting a prothrombotic state. PAI-1 is associated with metabolic syndrome, obesity, and cardiovascular risk. A key target in fibrinolysis regulation research and antithrombotic drug development.

Plasminogen (PLG) — The zymogen precursor of plasmin, the primary fibrinolytic protease. tPA and uPA cleave plasminogen to generate active plasmin, which degrades fibrin clots. Plasminogen deficiency causes ligneous conjunctivitis and impaired wound healing. Recombinant plasminogen proteins support functional fibrinolysis assays.

tPA (Tissue Plasminogen Activator / PLAT) — Serine protease that converts plasminogen to plasmin on the fibrin surface. Recombinant tPA (alteplase) is the standard thrombolytic therapy for acute ischemic stroke and ST-elevation MI. Anti-tPA antibodies support pharmacokinetic assays and ADA detection in clinical studies.
