Notch Signaling Pathway Research Reagents

The Notch pathway is an evolutionarily conserved cell-cell contact signaling mechanism that regulates cell fate decisions, proliferation, and differentiation. Ligand binding (DLL1/4, Jagged1/2) on a signal-sending cell triggers sequential proteolytic cleavage of the Notch receptor — first by ADAM10/17 metalloproteases, then by γ-secretase — releasing the Notch intracellular domain (NICD) for nuclear translocation and activation of HES/HEY family target genes. Aberrant Notch signaling is implicated in T-ALL, breast cancer, pancreatic cancer, and vascular development disorders.

Notch Receptors

Notch1 — The most extensively studied Notch receptor. Gain-of-function mutations in NOTCH1 are found in over 50% of T-ALL cases. Recombinant Notch1 extracellular domain proteins support ligand-binding studies and γ-secretase inhibitor screening.

Notch2 — Plays a critical role in marginal zone B cell development and bile duct morphogenesis. NOTCH2 mutations are associated with Hajdu-Cheney syndrome and splenic marginal zone lymphoma.

Notch3 — Predominantly expressed in vascular smooth muscle cells. NOTCH3 mutations cause CADASIL (cerebral autosomal dominant arteriopathy with subcortical infarcts), the most common hereditary cause of stroke and vascular dementia.

Notch Ligands

DLL4 (Delta-Like Ligand 4) — A key regulator of angiogenic sprouting and tip/stalk cell selection. DLL4-Notch1 signaling is a validated therapeutic target in tumor angiogenesis. Fc-tagged DLL4 proteins are recommended for plate-immobilized Notch activation assays.

DLL1 (Delta-Like Ligand 1) — Essential for somitogenesis, T cell lineage commitment in the thymus, and maintenance of intestinal stem cell homeostasis.

Jagged1 (JAG1) — A Serrate-family Notch ligand involved in Alagille syndrome (bile duct paucity), hematopoietic stem cell maintenance, and tumor-stromal Notch signaling. Jagged1-Notch2 interaction is critical for marginal zone B cell development.
