Pregnancy & Fertility Biomarkers

hCG (Human Chorionic Gonadotropin) — Glycoprotein hormone produced by placental trophoblasts, the definitive biomarker for pregnancy detection. hCG consists of an α-subunit (shared with FSH, LH, TSH) and a unique β-subunit (β-hCG) that confers specificity. Quantitative hCG assays monitor early pregnancy viability, ectopic pregnancy, and gestational trophoblastic disease. Recombinant hCG and β-hCG proteins serve as immunoassay calibrators; matched antibody pairs support ELISA, CLIA, and lateral flow rapid test development.

AMH (Anti-Müllerian Hormone / MIS) — A TGF-β superfamily glycoprotein produced by ovarian granulosa cells (females) and Sertoli cells (males). Serum AMH is the leading biomarker for ovarian reserve assessment in ART, predicting oocyte yield and guiding stimulation protocols. AMH is also used in polycystic ovary syndrome (PCOS) diagnosis and as a tumor marker for granulosa cell tumors.

LH (Luteinizing Hormone) — Pituitary gonadotropin that triggers ovulation and stimulates Leydig cell testosterone production. The LH surge is the physiological trigger for oocyte maturation and is detected by ovulation prediction kits. LH/FSH ratio is used in PCOS evaluation.

Activin (INHBA / INHBB) — TGF-β superfamily dimeric proteins (Activin A, Activin B, Activin AB) that stimulate FSH secretion from the pituitary and regulate folliculogenesis, embryogenesis, and erythropoiesis. Activin A is also an emerging biomarker for preeclampsia and is implicated in cachexia and muscle wasting via ActRII signaling.

Inhibin (INHA / INHBA / INHBB) — Heterodimeric glycoproteins (Inhibin A, Inhibin B) that suppress pituitary FSH secretion, forming a negative feedback loop with Activin. Inhibin B reflects Sertoli cell function in males and antral follicle count in females. Inhibin A is a component of the maternal serum quadruple screen for Down syndrome.

GnRH (Gonadotropin-Releasing Hormone) — Hypothalamic decapeptide that controls the HPG axis by stimulating pulsatile FSH and LH release. Estrogen Receptor (ESR1 / ESR2) — Nuclear hormone receptors that mediate estrogen signaling in reproductive tissues, bone, and the cardiovascular system. ERα (ESR1) is the primary therapeutic target in ER-positive breast cancer (tamoxifen, fulvestrant). ERβ (ESR2) modulates ERα activity and is implicated in endometriosis and ovarian function. Anti-ER antibodies are widely used in IHC for breast cancer subtyping.
