Dust Mite Allergen Research Reagents

House dust mites (HDM) are the most common perennial aeroallergen source globally, sensitizing over 1 billion people. Dermatophagoides pteronyssinus (Der p) and Dermatophagoides farinae (Der f) are the dominant species in temperate climates; Blomia tropicalis (Blo t) predominates in tropical regions. HDM sensitization is a major risk factor for allergic asthma development and persistence. Subcutaneous (SCIT) and sublingual (SLIT) immunotherapy targeting HDM allergens are guideline-recommended treatments for allergic rhinitis and mild-to-moderate asthma.

Key Dust Mite Allergens

Der p 1 / Der f 1 (Group 1 Cysteine Proteases) — The immunodominant HDM allergens with dual pathogenic activity: direct IgE-mediated sensitization plus enzymatic disruption of airway epithelial barrier (tight junction cleavage of occludin and claudin-1). Der p 1 and Der f 1 are 80% sequence-identical and broadly cross-reactive. Group 1 allergens are quantified as environmental exposure markers using monoclonal antibody-based ELISA.

Der p 2 / Der f 2 (Group 2 NPC2 Homologs) — Lipid-binding proteins structurally homologous to MD-2 (the LPS co-receptor of TLR4). Der p 2 can functionally replace MD-2 and activate innate immune signaling, providing a mechanistic link between HDM exposure and allergic airway inflammation. Der p 2 is the most frequently recognized HDM allergen in Southeast Asian populations.

Der p 23 (Peritrophin-Like Protein) — A recently characterized major HDM allergen found in mite fecal particles. Der p 23 sensitization prevalence reaches 74% in HDM-allergic patients, making it the third most important Der p allergen after groups 1 and 2. Its inclusion in CRD panels improves diagnostic sensitivity for HDM allergy.

Blo t 5 (Blomia tropicalis) — The major allergen of the tropical storage mite B. tropicalis, prevalent in Southeast Asia, South America, and coastal China. Blo t 5 shows limited cross-reactivity with Der p/f allergens, meaning HDM-SLIT formulated for Dermatophagoides may not adequately cover Blo t-sensitized patients in tropical regions.
