Bimagrumab + Semaglutide Dual-Target Mechanism: Myostatin/GLP-1 Pathway Research & Reagent Tools

Target 1: GLP-1 Receptor (Semaglutide)

Semaglutide is a long-acting GLP-1 receptor agonist that mimics the endogenous incretin hormone GLP-1. It suppresses appetite via hypothalamic GLP-1R signaling, delays gastric emptying, enhances glucose-dependent insulin secretion, and reduces glucagon release. The net effect is a sustained reduction in energy intake and body weight.

However, GLP-1-mediated weight loss is not fat-specific: multiple clinical studies report that 20–30% of total weight loss from GLP-1 RA monotherapy consists of lean mass (primarily skeletal muscle), which may impair long-term metabolic health and physical function. This limitation is the rationale for combining GLP-1 agonists with anabolic agents like bimagrumab.

Target 2: Activin Type II Receptor (Bimagrumab)

Bimagrumab is a monoclonal antibody that blocks both Activin Type IIA (ActRIIA) and Type IIB (ActRIIB) receptors, preventing signaling by TGF-β superfamily ligands including myostatin and activin A. These ligands are negative regulators of skeletal muscle growth: by blocking their receptors, bimagrumab promotes muscle protein synthesis, reduces fat deposition, and improves basal metabolic rate.
