Melanoma Research Reagents: BRAF, Immune Checkpoint & Melanocyte Markers

Melanoma has been at the forefront of immuno-oncology, serving as the proving ground for both CTLA-4 (ipilimumab, 2011) and PD-1 (pembrolizumab/nivolumab, 2014) checkpoint blockade, as well as the first LAG-3 combination (relatlimab + nivolumab, 2022). Key Targets and Biomarkers

BRAF V600E/K mutations occur in ~50% of cutaneous melanomas. Anti-BRAF V600E antibody (clone VE1) enables rapid IHC-based mutation screening on FFPE tissue. Wild-type BRAF melanomas are typically NRAS-mutant (~20%) or NF1-mutant (~15%).

Melanocyte lineage markers for IHC diagnosis: SOX10 (nuclear; most sensitive), S100 (nuclear + cytoplasmic; most commonly used), MITF (nuclear), Melan-A/MART-1 (cytoplasmic), HMB-45/gp100 (cytoplasmic). A panel approach (SOX10 + S100 + Melan-A) provides the highest diagnostic sensitivity. For guidance on building diagnostic IHC panels, see our IHC Panel Design guide.

Melanoma is highly immunogenic, with high tumor mutational burden (TMB) generating neoantigens that drive T cell responses. The tumor microenvironment in melanoma is a major focus of research, with TIL density and PD-L1 expression serving as predictive biomarkers for checkpoint immunotherapy response.
