Antibody-drug conjugates (ADCs) combine the targeting specificity of monoclonal antibodies with the cell-killing potency of cytotoxic payloads. With over 10 ADCs now approved globally and 100+ in clinical development, ADCs represent one of the fastest-growing modalities in oncology drug development. The success of an ADC program depends critically on target selection — the target antigen must be highly expressed on tumor cells, minimally expressed on normal tissue, and efficiently internalized upon antibody binding.

Research Applications for ADC Target Reagents

Target expression profiling by IHC. Evaluate target antigen expression levels and distribution in tumor tissue using anti-target antibodies on FFPE sections. Internalization assays. Use fluorophore-conjugated anti-target antibodies to measure receptor internalization kinetics by flow cytometry or confocal microscopy — a critical parameter for ADC payload delivery.

Binding kinetics (SPR/BLI). Use recombinant target proteins (ECD domains) as ligands on SPR chips or BLI biosensors to measure antibody-antigen binding affinity and kinetics for ADC candidate screening.

Cell-based potency assays. Recombinant target-expressing stable cell lines and recombinant proteins enable ADCC, CDC, and ADC-mediated cytotoxicity assays for lead candidate selection.

PK/ADA bioanalysis. Research biosimilar versions of approved ADC antibodies (e.g., trastuzumab, sacituzumab) serve as reference standards in pharmacokinetic and immunogenicity assays.

ADC targets are increasingly studied in combination with PD-1/PD-L1 checkpoint inhibitors in clinical trials, requiring multiplex reagents that span both target classes.

Q: What is the difference between the naked antibody and the ADC in terms of research reagents?
