Read at 450 nm with a reference wavelength of 630 nm (for TMB substrate). Read within 15 minutes of adding stop solution. Use a 4-parameter logistic (4PL) curve fit for standard curve calculation.
Sandwich ELISA uses two non-overlapping epitopes for high-sensitivity detection of proteins and cytokines (signal increases with concentration). Competitive ELISA works for small molecules or single-epitope targets (signal decreases with concentration).
