Selection & Best Practices

  1. Match Precisely: Same host species, isotype/subclass (e.g., mouse IgG1 κ), and the same conjugate as your primary; use the same concentration or mass per test.
  2. Fragment Choice: Choose F(ab')2 when Fc receptors cause background (macrophage-rich tissues, spleen, tumors).
  3. Cross-Adsorption: Select cross-adsorbed (or highly cross-adsorbed) for multi-species samples to reduce off-target binding.
  4. Flow Gating: Use FMO controls to set positive gates; use isotypes to estimate non-specific binding—not to define thresholds.
  5. Fix/Perm Compatibility: Verify tandem dye and fixation compatibility (paraformaldehyde, methanol, saponin) on the datasheet.

FAQs

Should I always use an isotype control?

Use FMO controls for gating in multicolor flow; isotypes help estimate non-specific binding from Fc/charge/hydrophobic interactions, especially in sticky tissues or when primaries are low-specificity.

How do I choose between whole IgG and F(ab')2?

Whole IgG maximizes signal; F(ab')2 lacks Fc to avoid FcR-mediated binding and often lowers background in IF/IHC and flow.

What “cross-adsorbed” means?

Antiserum is adsorbed against non-target species to reduce cross-reactivity; “highly cross-adsorbed” further minimizes off-target binding in multi-species samples.
