Selection Guide

  1. Use Case: Blocking (choose high-affinity, epitope-defined VHH) vs. imaging (small monovalent VHH or fluor-labeled).
  2. Valency/Fc: VHH-Fc or bivalent VHH increases avidity and half-life; monovalent reduces steric effects and improves penetration.
  3. Expression Host: E. coli for rapid screens; HEK/CHO for Fc fusions and glyco-dependent stability.
  4. Conjugation: Biotin/HRP for ELISA; Alexa 488/647 for IF/live-cell; keep free VHH for neutralization/MoA assays.
  5. Endotoxin & Monomer %: Check lot CoA (SEC-HPLC) for aggregation and endotoxin per sensitive cell assays.

What is a nanobody (VHH)?

A single-domain antibody derived from camelid heavy-chain-only antibodies; ~15 kDa, highly stable/soluble with excellent tissue penetration.

When should I choose VHH-Fc or bivalent formats?

Use VHH-Fc/bivalent for higher apparent affinity/half-life and plate/cell capture; choose monovalent for steric-sensitive epitopes or intracellular use.

Are these humanized?
