B7-H3 (CD276) is an immune checkpoint molecule of the B7 family that is broadly overexpressed on solid tumors — including NSCLC, prostate, ovarian, and pediatric cancers — while showing limited expression on normal tissues. This tumor-selective expression profile makes B7-H3 an attractive target for antibody-drug conjugates (ADCs), CAR-T therapy, and checkpoint immunotherapy. Several anti-B7-H3 therapeutics are in advanced clinical development, including Omburtamab (8H9), Vobramitamab duocarmazine (MGC018), and Enoblituzumab (MGA271). abinScience offers recombinant B7-H3 proteins, anti-B7-H3 antibodies, biosimilar reference standards, and ELISA kits.
B7-H3 Biology and Therapeutic Relevance
B7-H3 is a type I transmembrane glycoprotein with two structural isoforms: 2Ig-B7-H3 (found in mice, containing one IgV-IgC domain pair) and 4Ig-B7-H3 (the dominant human isoform, containing a tandem duplication of the IgV-IgC domains). The 4Ig form is ~100 kDa and is the primary target of therapeutic antibodies in clinical development.
Unlike PD-L1, whose receptor (PD-1) is well characterized, the cognate receptor for B7-H3 on T cells remains unidentified. B7-H3 has demonstrated both co-stimulatory and co-inhibitory functions depending on the experimental context. In the tumor microenvironment, B7-H3 overexpression correlates with reduced T-cell infiltration, increased tumor invasion, and poor prognosis. Beyond immune modulation, B7-H3 also promotes tumor cell migration, invasion, and chemoresistance through non-immune signaling pathways (JAK2/STAT3, PI3K/AKT).
Key Research Applications
ADC & Bispecific Development
Recombinant B7-H3 proteins (4Ig and 2Ig isoforms) for anti-B7-H3 antibody screening, ADC payload conjugation assessment, and bispecific T-cell engager development targeting B7-H3 × CD3.
CAR-T Target Validation
B7-H3 proteins and B7-H3-expressing stable cell lines for CAR-T construct screening, cytotoxicity assays, and in vitro potency testing. B7-H3's broad tumor expression and limited normal tissue expression make it a preferred solid tumor CAR-T target.
IHC & Tumor Expression Profiling
Anti-B7-H3 antibodies validated for IHC on FFPE tumor sections. B7-H3 IHC is increasingly used to stratify patients for anti-B7-H3 clinical trials and to characterize tumor microenvironment composition.
PK/ADA Assays
Anti-B7-H3 antibody pairs and B7-H3 calibrator proteins for sandwich ELISA-based pharmacokinetic and anti-drug antibody assay development supporting anti-B7-H3 clinical programs.

B7-H3 FAQs
What is the difference between 2Ig-B7-H3 and 4Ig-B7-H3?
The 2Ig isoform contains one IgV-IgC domain pair and is the predominant form in mice. The 4Ig isoform contains a tandem duplication (two IgV-IgC pairs) and is the dominant form in humans (~100 kDa vs ~45 kDa). Most therapeutic anti-B7-H3 antibodies in clinical trials are raised against the 4Ig human isoform. When selecting reagents for preclinical work, ensure the protein isoform matches your experimental species — use 4Ig for human studies and 2Ig for mouse syngeneic tumor models.
Why is B7-H3 considered a better ADC target than PD-L1?
B7-H3 has several properties that favor ADC development: (1) high and homogeneous tumor surface expression across many solid tumor types, (2) limited expression on normal tissues (reducing off-target toxicity), (3) efficient receptor-mediated internalization upon antibody binding (essential for intracellular payload delivery), and (4) expression on tumor vasculature in addition to tumor cells (dual targeting effect). PD-L1, while broadly expressed on tumors, is also expressed on normal immune cells and tissues, increasing the risk of on-target off-tumor toxicity for ADC approaches.
What is the receptor for B7-H3?
The cognate receptor for B7-H3 on T cells has not been definitively identified, which is unusual for a B7 family member. TREM-like transcript 2 (TLT-2 / TREML2) was initially proposed as a receptor but this finding has been disputed. Several other candidate receptors have been suggested, including interleukin-20 receptor subunit alpha (IL-20Rα). This unresolved receptor biology means that most therapeutic approaches to B7-H3 (ADCs, CAR-T) exploit its tumor expression pattern rather than its signaling function.
Which tumors express B7-H3 most strongly?
B7-H3 is overexpressed in a wide range of solid tumors, with particularly high expression reported in prostate cancer, NSCLC, ovarian cancer, neuroblastoma, glioblastoma, triple-negative breast cancer, and pancreatic ductal adenocarcinoma. In pediatric cancers, B7-H3 expression is especially prominent — Omburtamab (anti-B7-H3 8H9 antibody) was developed for CNS neuroblastoma. B7-H3 is also expressed on tumor-associated vasculature, which contributes to the therapeutic window.
How should I detect B7-H3 by IHC?
For FFPE sections, use an anti-B7-H3 monoclonal antibody with heat-induced epitope retrieval (citrate pH 6.0 or Tris-EDTA pH 9.0 — optimize for your tissue type). B7-H3 staining is typically membranous and cytoplasmic. Include a known B7-H3-positive tumor section (prostate or neuroblastoma) as positive control. For dual staining with immune cell markers, combine with CD8 or CD68 to correlate B7-H3 expression with T-cell or macrophage infiltration.
