| Background | T-cell surface glycoprotein CD3 epsilon chain (T3E,T-cell surface antigen T3/Leu-4 epsilon chain,CD3e,T-cell surface glycoprotein CD3 epsilon chain) is a ~23 kDa protein. Part of the TCR-CD3 complex present on T-lymphocyte cell surface that plays an essential role in adaptive immune response. When antigen presenting cells (APCs) activate T-cell receptor (TCR), TCR-mediated signals are transmitted across the cell membrane by the CD3 chains CD3D, CD3E, CD3G and CD247/CD3Z. All CD3 chains contain immunoreceptor tyrosine-based activation motifs (ITAMs) in their cytoplasmic domain. Upon TCR engagement, these motifs become phosphorylated by Src family protein tyrosine kinases LCK and FYN, resulting in the activation of downstream signaling pathways. CD3E ITAM phosphorylation creates docking sites for the protein kinase ZAP70 leading to ZAP70 phosphorylation and its conversion into a catalytically active enzyme. 1. Gimferrer, I. et al. Western blot analysis was performed using anti-CD3 epsilon monoclonal antibody at 1 ug/mL on various samples.
Lane 1: Jurkat cell lysate

Jurkat cells were fixed with 4% paraformaldehyde for 20 minutes, followed by blocking with 5% goat serum for 1 hour. The cells were then incubated with a primary antibody targeting CD3 epsilon (HY057073) at a concentration of 20 ug/ml overnight at 4°C. Afterward, the cells were incubated with a secondary antibody, Goat Anti-Rabbit IgG (Alexa Fluor-488) at a concentration of 6.5 ug/ml for 50 minutes at room temperature. The localization of CD3 epsilon was visualized (shown in green), and nuclear DNA was stained with DAPI (shown in blue).

Flow-cytometry using anti-Human CD3 epsilon antibody. Jurkat cells were stained with an irrelevant antibody (Blue Histogram) or an anti-Human CD3 epsilon monoclonal antibody (Catalog HY057073, Yellow Histogram) at a concentration of 5 µg/ml for 30 mins at RT. After washing, bound antibody was detected using a Goat Anti-Rabbit IgG H&L Polyclonal Antibody, FITC (abinScience:TF690414) and cells analysed on a NovoCyte Flow Cytometer.
