| Background | ADP-ribosyl cyclase/cyclic ADP-ribose hydrolase 1 (CD38/ADPRC 1) is a ~34 kDa protein. Multifunctional transmembrane glycoprotein able to exert enzymatic activities and also to mobilize calcium, to transduce signals, to adhere to hyaluronan and to other ligands. Synthesizes cyclic ADP-ribose (cADPR), a second messenger for glucose-induced insulin secretion. Synthesizes the Ca(2+) mobilizer nicotinate-adenine dinucleotide phosphate, NAADP(+), from 2'-phospho-cADPR and nicotinic acid, as well as from NADP(+) and nicotinic acid. At both pH 5.0 and pH 7.4 preferentially transforms 2'-phospho-cADPR into NAADP(+), while preferentially cleaving NADP(+) to cADPR and ADPRP rather than into NADDP(+). Has cADPR hydrolase activity. CD38 is the therapeutic target of daratumumab (Darzalex). 1. Tohgo, A. et al. (1994) The Journal of biological chemistry 269, 28555-7. PMID: 7961800 2. Takasawa, S. et al. Raji cells were fixed with 4% paraformaldehyde for 20 minutes, followed by blocking with 5% goat serum for 1 hour. The cells were then incubated with a primary antibody targeting CD38 (HB691083) at a concentration of 20 ug/ml overnight at 4°C. Afterward, the cells were incubated with a secondary antibody, Goat Anti-Rabbit IgG (Alexa Fluor-488) at a concentration of 6.5 ug/ml for 50 minutes at room temperature. The localization of CD38 was visualized (shown in green), and nuclear DNA was stained with DAPI (shown in blue).

Flow-cytometry using anti-Human CD38 antibody. RPMI-8226 cells were stained with an irrelevant antibody (Blue Histogram) or an anti-Human CD38 monoclonal antibody (Catalog HB691083, Yellow Histogram) at a concentration of 5 µg/ml for 30 mins at RT. After washing, bound antibody was detected using a Goat Anti-Rabbit IgG H&L Polyclonal Antibody, FITC (abinScience:TF690414) and cells analysed on a NovoCyte Flow Cytometer.
