| Background | Epidermal growth factor receptor (EGFR/ERBB1) is a ~134 kDa protein. Receptor tyrosine kinase binding ligands of the EGF family and activating several signaling cascades to convert extracellular cues into appropriate cellular responses. Known ligands include EGF, TGFA/TGF-alpha, AREG, epigen/EPGN, BTC/betacellulin, epiregulin/EREG and HBEGF/heparin-binding EGF. Ligand binding triggers receptor homo- and/or heterodimerization and autophosphorylation on key cytoplasmic residues. The phosphorylated receptor recruits adapter proteins like GRB2 which in turn activates complex downstream signaling cascades. Activates at least 4 major downstream signaling cascades including the RAS-RAF-MEK-ERK, PI3 kinase-AKT, PLCgamma-PKC and STATs modules. EGFR is the therapeutic target of cetuximab (Erbitux) and panitumumab (Vectibix). 1. Arcaro, A. et al. (2000) Molecular and cellular biology 20, 3817-30. PMID: 10805725 2. Runkle, KB. et al. | Note | For flow cytometric staining, the suggested use of this reagent is 0.5 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application. For research use only. |

Flow-cytometry using PE anti-human EGFR antibody. A431 cells were stained with an irrelevant antibody (Blue Histogram) or an PE anti-human EGFR monoclonal antibody (Catalog: HF004327, Yellow Histogram) at a concentration of 5 µg/ml for 30 mins at RT. After washing, and cells analysed on a NovoCyte Flow Cytometer.
