| Background | Epidermal growth factor receptor (EGFR/ERBB1) is a ~134 kDa protein. Receptor tyrosine kinase binding ligands of the EGF family and activating several signaling cascades to convert extracellular cues into appropriate cellular responses. Known ligands include EGF, TGFA/TGF-alpha, AREG, epigen/EPGN, BTC/betacellulin, epiregulin/EREG and HBEGF/heparin-binding EGF. Ligand binding triggers receptor homo- and/or heterodimerization and autophosphorylation on key cytoplasmic residues. The phosphorylated receptor recruits adapter proteins like GRB2 which in turn activates complex downstream signaling cascades. Activates at least 4 major downstream signaling cascades including the RAS-RAF-MEK-ERK, PI3 kinase-AKT, PLCgamma-PKC and STATs modules. EGFR is the therapeutic target of cetuximab (Erbitux) and panitumumab (Vectibix). 1. Arcaro, A. et al. (2000) Molecular and cellular biology 20, 3817-30. PMID: 10805725 2. Runkle, KB. et al. Flow-cytometry using APC anti-human EGFR antibody. A431 cells were stained with an irrelevant antibody (Blue Histogram) or an APC anti-human EGFR monoclonal antibody (Catalog: HF004056, Yellow Histogram) at a concentration of 5 µg/ml for 30 mins at RT. After washing, and cells analysed on a NovoCyte Flow Cytometer.

Flow-cytometry using PE anti-human EGFR antibody. A431 cells were stained with an irrelevant antibody (Blue Histogram) or an PE anti-human EGFR monoclonal antibody (Catalog: HF004056, Yellow Histogram) at a concentration of 5 µg/ml for 30 mins at RT. After washing, and cells analysed on a NovoCyte Flow Cytometer.

Flow-cytometry using FITC anti-human EGFR antibody. A431 cells were stained with an irrelevant antibody (Blue Histogram) or an FITC anti-human EGFR monoclonal antibody (Catalog: HF004056, Yellow Histogram) at a concentration of 5 µg/ml for 30 mins at RT. After washing, and cells analysed on a NovoCyte Flow Cytometer.

Flow-cytometry using anti-human EGFR antibody. Untransfected cells (blue Histogram) and Transfected cells (Yellow Histogram) were stained with an anti-human EGFR monoclonal antibody (Catalog: HF004056) at a concentration of 5 µg/ml for 30 mins at RT. After washing, bound antibody was detected using a Goat Anti-Human IgG H&L Polyclonal Antibody, FITC (abinScience: HF690414) and cells analysed on a NovoCyte Flow Cytometer.
