Biological activity AO-176 shows EC 50 values of 130 ng/mL (OV90), 390 ng/mL (HCC827), 649 ng/mL (SNU-1), 250 ng/mL (OV10-315), 2700 ng/mL (MDA-MB-231), 390 ng/mL (Jurkat) and 1910 ng/mL (Raji) [1]. AO-176 (10 ug/mL, 24 h) induces death of Jurkat and OV90 cells [1]. AO-176 (0.017-1000 ug/mL) binds negligibly to RBCs and does not cause agglutination of washed human RBCs [1][3]. AO-176 (10-30 ug/mL, 24 h) has no killing effect on normal cell and the activated T cells [1]. AO-176 (0.001-100 ug/mL, 90 min) directly inhibits the binding of SIRPα to CD47 on Jurkat cells with an IC 50 of 0.78 to 0.87 ug/mL [1]. AO-176 (0.01-10 ug/mL, 2 h) promotes the phagocytosis of both Jurkat, Raji, OV90, Detroit 562, and FaDu cells in a concentration-dependent manner [1][2][4]. AO-176 (1-25 mg/kg, i.v., weekly for 4 weeks) significantly suppresses the tumor growth in Raji lymphoma xenograft mice models [1]. AO-176 (10-25 mg/kg, i.p., weekly or 5 times/weeks for 5-6 weeks) significantly suppresses the tumor growth in MDA-MB-23, SNU-1 and OV90 xenograft mice models [1]. AO-176 (3-25 mg/kg) exerts anti-tumor activity in RPMI-8226 and NCI-H929 xenografted mice models [3][4]. .

Background

Leukocyte surface antigen CD47 (MER6) is a ~35 kDa protein. Adhesive protein that mediates cell-to-cell interactions. Acts as a receptor for thrombospondin THBS1 and as modulator of integrin signaling through the activation of heterotrimeric G proteins. Involved in signal transduction, cardiovascular homeostasis, inflammation, apoptosis, angiogenesis, cellular self-renewal, and immunoregulation. Plays a role in modulating pulmonary endothelin EDN1 signaling. Modulates nitrous oxide (NO) signaling, in response to THBS1, hence playing a role as a pressor agent, supporting blood pressure. CD47 is the therapeutic target of magrolimab (clinical).
