Background	Hematopoietic progenitor cell antigen CD34 is a ~40 kDa protein. Possible adhesion molecule with a role in early hematopoiesis by mediating the attachment of stem cells to the bone marrow extracellular matrix or directly to stromal cells. Could act as a scaffold for the attachment of lineage specific glycans, allowing stem cells to bind to lectins expressed by stromal cells or other marrow components. Presents carbohydrate ligands to selectins.

Flow Cytometry

Flow-cytometry using anti-human CD34 antibody. Human MOLT-4 cell line were stained with an irrelevant antibody (Green Histogram) or an anti-human CD34 antibody monoclonal antibody (Catalog # HB766107 , Red Histogram) at a concentration of 5 µg/ml for 30 mins at RT. After washing, bound antibody was detected using a CoraLite488 conjugated goat anti-mouse antibody (Catalog # MF690314) and cells analysed on a NovoCyte Flow Cytometer.

KG-1 cells were stained with an irrelevant antibody (Blue Histogram) or an anti-human CD34 monoclonal antibody (Catalog HB766107, Yellow Histogram) at a concentration of 5 µg/ml for 30 mins at RT. After washing, bound antibody was detected using a Goat Anti-Mouse IgG H&L Polyclonal Antibody, FITC (abinScience: MF690414) and cells analysed on a NovoCyte Flow Cytometer.
