Background

Outer capsid protein sigma-3 (Sigma3/S4) is a ~41 kDa protein. Outer layer capsid protein that stimulates translation by blocking the activation of the dsRNA-dependent protein kinase EIF2AK2/PKR, thereby inhibiting the host interferon response. Sigma3 prevents the activation of EIF2AK2 by competing with the kinase for dsRNA-binding. During virus assembly, the inner layer proteins lambda-1 and the turret protein lambda-2 form the star core, followed by the stepwise attachment of sigma-2 that completes the core and mu1/sigma3 that complete the outer layer. Binds to the host neuronal receptor RTN4R for viral entry into host neural cells.
