| Background | Dual specificity protein kinase TTK is a ~97 kDa protein. Involved in mitotic spindle assembly checkpoint signaling, a process that delays anaphase until chromosomes are bioriented on the spindle, and in the repair of incorrect mitotic kinetochore-spindle microtubule attachments. Phosphorylates MAD1L1 to promote the mitotic spindle assembly checkpoint. Phosphorylates CDCA8/Borealin leading to enhanced AURKB activity at the kinetochore. Phosphorylates SKA3 at 'Ser-34' leading to dissociation of the SKA complex from microtubules and destabilization of microtubule-kinetochore attachments. Phosphorylates KNL1, KNTC1 and autophosphorylates. 1. Jelluma, N. et al. (2008) Cell 132, 233-46. PMID: 18243099 2. Maciejowski, J. et al. (2017) Developmental cell 41, 143-156.e6. PMID: 28441529 3.
