Biological activity HFB101110 (0.01-10 nM) dose-dependently inhibits hCCL1-induced chemotaxis of CCR8 + cells [1]. HFB101110 (10 ug/mL) completely inhibits hCCL1-induced calcium flux in CCR8 + cells [1]. HFB101110 (0.3-300 ug/mL) does not induce significant cytokine release from healthy human PBMCs at concentrations up to 300 ug/mL [1]. HFB101110 exerts potent single-agent antitumor activity in a human CCR8 knock-in mouse model via depletion of intratumoral regulatory T cells [2]. HFB101110 is well-tolerated in wild-type mice [2]. HFB101110 is well-tolerated in cynomolgus monkeys [2].

Background

C-C chemokine receptor type 8 (CD198/CCR8) is a ~40 kDa protein. G protein-coupled receptor that can bind a variety of chemokines, such as CCL1, CCL8, CCL16, CCL18. Regulates monocyte and eosinophil chemotaxis. Undergoes internalization upon CCL18 binding, leading to induced migration and calcium flux of highly polarized Th2 cells. In microglial cells, promotes phagocytosis with CCL18 through NF-kappa-B and Src signaling pathways. Stimulation of the CCL1-CCR8 signaling axis protects the gut from acute intestinal damage.
