| Biological activity | ATG-031 potently binds to CD24-postive tumor cells, while showing no binding with parental HEK293 cells or hRBC [1]. ATG-031 blocks the interaction between CD24 and Siglec-10 and induces potent macrophage-dependent tumor cell phagocytosis [1]. ATG-031 induces a potent antibody-dependent cellular cytotoxicity against HT-29 tumor cells with an EC 50 of 0.27 nM [3]. ATG-031 (1-3 mg/kg, i.v., biweekly) significantly inhibits tumor growth and induces tumor regression in MC38-hCD24 mouse model [1]. ATG-031 exhibits potent antitumor activity in vivo as a single agent in syngeneic models such as MC38, CT26, and 4T1 [3]. ATG-031 demonstrates in vivo synergism with chemotherapies and checkpoint inhibitors [3]. ATG-031 (3 mg/kg, i.v., 2 doses) significantly increases T cell-associated immune response at the cellular and single-cell transcriptome levels in C57BL/6 mice bearing MC38-hCD24 tumors [3]. |

| Background | Signal transducer CD24 is a ~8 kDa protein. May have a pivotal role in cell differentiation of different cell types. Signaling could be triggered by the binding of a lectin-like ligand to the CD24 carbohydrates, and transduced by the release of second messengers derived from the GPI-anchor. Modulates B-cell activation responses. Promotes AG-dependent proliferation of B-cells, and prevents their terminal differentiation into antibody-forming cells.
