Biological activity BI-836858 (10 ug/mL; 30 min) mediates specific ADCC activity against primary AML blasts [1]. BI-836858 (2 ug/mL; 4 h) mediates NK cell-dependent ADCC activity against CD33-positive HL60 cells [2]. BI-836858 (2 ug/mL; 48 h) blocks the CD33-mediated inhibitory signaling pathway in MDS bone marrow cells and reduces the expression of IL-10 and TGFβ [2]. BI-836858 (2 ug/mL) specifically blocks CD33 (S100A9)-mediated ROS production in U937 cells, healthy PBMCs, and primary MDS BM HSPCs [2]. BI-836858 (2 ug/mL; 14 days) restores the hematopoietic colony-forming capacity in ex vivo cultures of primary MDS bone marrow specimens [2].

Background

Myeloid cell surface antigen CD33 is a ~39 kDa protein. Sialic-acid-binding immunoglobulin-like lectin (Siglec) that plays a role in mediating cell-cell interactions and in maintaining immune cells in a resting state. Preferentially recognizes and binds alpha-2,3- and more avidly alpha-2,6-linked sialic acid-bearing glycans. Upon engagement of ligands such as C1q or syalylated glycoproteins, two immunoreceptor tyrosine-based inhibitory motifs (ITIMs) located in CD33 cytoplasmic tail are phosphorylated by Src-like kinases such as LCK. These phosphorylations provide docking sites for the recruitment and activation of protein-tyrosine phosphatases PTPN6/SHP-1 and PTPN11/SHP-2. In turn, these phosphatases regulate downstream pathways through dephosphorylation of signaling molecules. CD33 is the therapeutic target of gemtuzumab ozogamicin (Mylotarg).
