Biological activity OM-RCA-01 binds to FGFR1 with high affinity (K d = 1.59 nM) and inhibits its in vitro kinase activity [1]. OM-RCA-01 (1-100 ug/mL, 3 h) potently inhibits FGFR1 phosphorylation and cell proliferation in FGFR1-overexpressing human lung cancer cell line A549 (GI 50 = 9.69 ug/mL), but exerts no significant effect on FGFR1-low-expressing human melanoma cell line Mel Kor [1]. OM-RCA-01 (0.5-10 nmol/L; 5 days) enhances Nivolumab -induced IFN-γ release and T-cell proliferation in a human allogeneic MLR assay, whereas it exhibits minimal effects when used alone when combined with Nivolumab () [1]. OM-RCA-01 (1 ug/mL; 4 days) increases Nivolumab -induced IL-2 secretion by 29%-74% in human peripheral blood mononuclear cells (PBMCs) stimulated with Staphylococcal enterotoxin B (SEB) () [1]. Background

Fibroblast growth factor receptor 1 (CD331/FGFR1) is a ~91 kDa protein. Tyrosine-protein kinase that acts as a cell-surface receptor for fibroblast growth factors and plays an essential role in the regulation of embryonic development, cell proliferation, differentiation and migration. Required for normal mesoderm patterning and correct axial organization during embryonic development, normal skeletogenesis and normal development of the gonadotropin-releasing hormone (GnRH) neuronal system. Phosphorylates PLCG1, FRS2, GAB1 and SHB. Ligand binding leads to the activation of several signaling cascades. Activation of PLCG1 leads to the production of the cellular signaling molecules diacylglycerol and inositol 1,4,5-trisphosphate.

    Flow CytoMetry

    Flow-cytometry using PE anti-human CD331 antibody. MG-63 cells were fixed and permeabilized, then stained with an irrelevant antibody (Blue Histogram) or an PE anti-human CD331 monoclonal antibody (Catalog: HY105026, Yellow Histogram) at a concentration of 5 µg/ml for 30 mins at RT. After washing, cells analysed on a NovoCyte Flow Cytometer.
