Biological activity NC762 (0.01-100 nM) binds to B7-H4 with an EC 50 of 1.12 nM in 624Mel.B7H4 cells and EC 50 of 0.976 nM in SKBR3 cells [2]. NC762 (0.1 ng/mL-100 ug/mL) binds to B7-H4 in human 293T cells but not mouse B7-H4 [2]. NC762 (0.1 pM-1000 nM) suggests increased binding to CD16a compared to V5.hlgG 1 [2]. NC762 (0.1-100 pM, 5 h) exhibits ADCC activity in 624Mel.B7H4 and SKBR3 cultured with effector Jurkat reporter cell line [2]. through binding to tumor-expressing B7-H4. NC762 can be studies in research on cancer such as advanced or metastatic solid tumors [1].

Background

V-set domain-containing T-cell activation inhibitor 1 (VTCN1/B7-H4) is a ~30 kDa protein. Negatively regulates T-cell-mediated immune response by inhibiting T-cell activation, proliferation, cytokine production and development of cytotoxicity. When expressed on the cell surface of tumor macrophages, plays an important role, together with regulatory T-cells (Treg), in the suppression of tumor-associated antigen-specific T-cell immunity. Involved in promoting epithelial cell transformation.
