| Biological activity | IPH4301 induces death of tumor cells expressing MICA/B in vitro via antibody-dependent cellular cytotoxicity and antibody-dependent cellular phagocytosis [1]. IPH4301 blocks the binding of MICA/B to NKG2D, and restores the expression and function of NKG2D on primary human NK cells and T cells [1]. IPH4301 reverses the immunosuppressive effect induced by M2 macrophages and restores the ADCC activity of autologous NK cells against MICA-expressing tumor cell lines [1]. IPH4301-derived PEG-G5.NH2-FITC-DOTA(Gd)-Monalizumab/IPH4301 nanoprobe can efficiently bind to NK-92MI cells. Its binding efficiency and in vitro MRI signal increase with elevated concentrations; besides, it upregulates IFN-γ secretion of NK-92MI cells, enhances the cytotoxicity against MDA-MB-231 triple-negative breast cancer cells and induces tumor cell apoptosis [2]. | ||||

| Background | MHC class I polypeptide-related sequence A (MICA/MICB) is a ~42 kDa protein. Widely expressed membrane-bound protein which acts as a ligand to stimulate an activating receptor KLRK1/NKG2D, expressed on the surface of essentially all human natural killer (NK), gammadelta T and CD8 alphabeta T-cells. Up-regulated in stressed conditions, such as viral and bacterial infections or DNA damage response, serves as signal of cellular stress, and engagement of KLRK1/NKG2D by MICA triggers NK-cells resulting in a range of immune effector functions, such as cytotoxicity and cytokine production. 1.
