Beta-Actin (ACTB) Antibodies & Proteins — Western Blot Loading Control

Beta-Actin (ACTB) is the second most popular loading control in western blot after GAPDH — and in some labs, it is the first choice. As a cytoskeletal housekeeping protein expressed at high, stable levels in virtually all mammalian cell types, Beta-Actin provides a reliable normalization reference at ~42 kDa, conveniently positioned in the middle of most SDS-PAGE gels where resolution is best.

When to Choose Beta-Actin Over GAPDH

Your target is near 36 kDa. GAPDH runs at ~36 kDa. If your protein of interest is in the 30–40 kDa range, the GAPDH band may overlap or complicate stripping and reprobing. Beta-Actin at ~42 kDa gives you more separation.

You are studying glycolysis, hypoxia, or metabolic stress. GAPDH is a glycolytic enzyme whose expression changes under these conditions. Beta-Actin is a structural protein and is generally more stable in metabolic perturbation experiments.

You need IP-validated loading control detection. Our anti-Beta-Actin nanobody (SAA series) is validated for IP — useful when you need to immunoprecipitate actin complexes or confirm Beta-Actin levels in pulldown experiments. GAPDH antibodies are typically not IP-validated.

Avoid Beta-Actin when: studying cytoskeletal dynamics, cell migration, or actin polymerization — Beta-Actin expression and distribution change in these contexts. Use Histone H3 (nuclear fraction) or Vinculin (~124 kDa, avoids the 30–50 kDa range entirely) as alternatives.

Beta-Actin Western Blot: Quick Reference

Suggested starting conditions. Optimal dilutions should be determined by the end user for each specific sample type.

Molecular weight ~42 kDa. Clean single band on standard 10–12% SDS-PAGE gels.
Suggested WB dilution Recombinant (SAA0339): 1:5000–1:20000. Polyclonal: 1:500–1:2000. Start high — Beta-Actin is abundant and overexposure is common.
Positive controls Any mammalian cell line lysate (HeLa, HEK293, NIH/3T3, Jurkat). Beta-Actin is ubiquitous.
Nanobody advantage The VHH nanobody (HX034023) is only ~15 kDa — it avoids the IgG heavy chain (~50 kDa) and light chain (~25 kDa) bands that can interfere with reprobing. Also validated for IP.
Beta vs Gamma Actin Beta-Actin (ACTB, 42 kDa) and Gamma-Actin (ACTG1, 42 kDa) co-migrate on SDS-PAGE. If you need to distinguish them, use isoform-specific antibodies. Our anti-ACTG1 polyclonal detects Gamma-Actin specifically.
