Getah virus (GETV) is an enveloped, positive-sense single-stranded RNA alphavirus of the family Togaviridae (~11.7 kb genome), transmitted by Culex and Aedes mosquitoes. GETV causes febrile illness, limb edema, rash, and reproductive failure in horses and swine — with major outbreaks reported in Japan, China, India, and Southeast Asia. GETV is closely related to other Old World alphaviruses including Sindbis virus (SINV), chikungunya virus (CHIKV), and Ross River virus (RRV). In recent years, GETV has gained increased attention due to expanding geographic range, detection in multiple livestock species (horses, pigs, cattle, foxes), and serological evidence of human infection in endemic areas. The virion encodes structural proteins capsid (C), envelope glycoproteins E1 (class II fusion) and E2 (receptor binding, primary neutralization target), plus four non-structural proteins (nsP1–nsP4). E2 determines receptor tropism and is the immunodominant target for neutralizing antibodies.

Spike Glycoprotein E2 — Receptor Binding & Neutralization Target
E2 is the major surface glycoprotein that mediates receptor attachment and is the primary target for virus-neutralizing antibodies. E2 forms a heterodimer with E1 and assembles into trimeric spikes on the virion surface. Anti-E2 antibodies block initial receptor binding and are the most potent neutralizers. E2 is also the key antigen for serological surveillance ELISA development — critical for GETV outbreak monitoring in equine and swine populations. Spike Glycoprotein E1 — Class II Fusion Protein
E1 is the class II fusion glycoprotein that mediates low-pH-triggered membrane fusion in endosomes. E1 is shielded by E2 in the prefusion state and is more conserved across alphavirus species than E2, making anti-E1 reagents useful for cross-alphavirus comparative studies. Which protein is better for GETV serosurvey ELISA — E1 or E2?
E2 (VK157022) is recommended for species-specific GETV serology — E2 contains the hypervariable neutralizing epitopes that distinguish GETV from related alphaviruses. E1 is more conserved and may cross-react with SINV, CHIKV, and RRV sera. Use E2 for GETV-specific detection and E1 for pan-alphavirus screening.
Do GETV reagents cross-react with CHIKV or SINV?
E2 shares limited amino acid identity with CHIKV E2 (~55%) and SINV E2 (~60%), so E2-based reagents are relatively GETV-specific. E1 is more conserved (~70–75% identity with SINV E1), and our anti-E1 pAb may show partial cross-reactivity with related Old World alphaviruses. Why is GETV getting more research attention?
GETV has been expanding geographically and into new host species. Large outbreaks in Chinese pig farms (2017–present) and Japanese racehorses, combined with serological evidence of human exposure, have raised concern about GETV as an emerging zoonotic threat. There are currently no approved GETV vaccines for any species, and diagnostic tools remain limited — our E1/E2 proteins and antibodies fill a critical reagent gap.
