| Background | L-selectin (CD62L/SELL) is a ~42 kDa protein. Calcium-dependent lectin that mediates cell adhesion by binding to glycoproteins on neighboring cells. Mediates the adherence of lymphocytes to endothelial cells of high endothelial venules in peripheral lymph nodes. Promotes initial tethering and rolling of leukocytes in endothelia. 1. Jurkat cells were fixed with 4% paraformaldehyde for 20 minutes, followed by blocking with 5% goat serum for 1 hour. The cells were then incubated with a primary antibody targeting L-Selectin/CD62L (HB644013) at a concentration of 20 ug/ml overnight at 4°C. Afterward, the cells were incubated with a secondary antibody, Goat Anti-Rabbit IgG (Alexa Fluor-488) at a concentration of 6.5 ug/ml for 50 minutes at room temperature. The localization of L-Selectin/CD62L was visualized (shown in green), and nuclear DNA was stained with DAPI (shown in blue).

Flow-cytometry using anti-Human CD62L antibody. Loucy cells were stained with an irrelevant antibody (Blue Histogram) or an anti-Human CD62L monoclonal antibody (Catalog HB644013, Yellow Histogram) at a concentration of 5 µg/ml for 30 mins at RT. After washing, bound antibody was detected using a Goat Anti-Rabbit IgG H&L Polyclonal Antibody, FITC (abinScience:TF690414) and cells analysed on a NovoCyte Flow Cytometer.
