Background

Serine/threonine-protein kinase B-raf (BRAF) is a ~84 kDa protein. Protein kinase involved in the transduction of mitogenic signals from the cell membrane to the nucleus (Probable). Phosphorylates MAP2K1, and thereby activates the MAP kinase signal transduction pathway. Phosphorylates PFKFB2. May play a role in the postsynaptic responses of hippocampal neurons.

    Immunofluorescence

HeLa cells were fixed with 4% paraformaldehyde for 20 minutes, followed by blocking with 5% goat serum for 1 hour. The cells were then incubated with a primary antibody targeting B-Raf (HB617043) at a concentration of 20 ug/ml overnight at 4°C. Afterward, the cells were incubated with a secondary antibody, Goat Anti-Rabbit IgG (Alexa Fluor-488) at a concentration of 6.5 ug/ml for 50 minutes at room temperature. The localization of B-Raf was visualized (shown in green), and nuclear DNA was stained with DAPI (shown in blue).
