| Background | HEK293T is a highly transfectable derivative of the human embryonic kidney 293 (HEK293) cell line that stably expresses the SV40 large T antigen. The presence of the T antigen allows episomal replication of transfected plasmids containing the SV40 origin of replication, leading to amplification of the plasmid and extended temporal expression of the desired gene products. HEK293T cells are widely used for transient protein expression, lentiviral and retroviral packaging, and as host cells for generating stable cell lines overexpressing specific target genes. They grow as an adherent monolayer and are readily maintained in standard culture conditions. |
