| Background | Chondroitin sulfate proteoglycan 4 (CSPG4/HMW-MAA) is a ~250 kDa protein. Proteoglycan playing a role in cell proliferation and migration which stimulates endothelial cells motility during microvascular morphogenesis. May also inhibit neurite outgrowth and growth cone collapse during axon regeneration. Cell surface receptor for collagen alpha 2(VI) which may confer cells ability to migrate on that substrate. Binds through its extracellular N-terminus growth factors, extracellular matrix proteases modulating their activity. May regulate MPP16-dependent degradation and invasion of type I collagen participating in melanoma cells invasion properties. | ||||||||

U2OS cells were fixed with 4% paraformaldehyde for 20 minutes, followed by blocking with 5% goat serum for 1 hour. The cells were then incubated with a primary antibody targeting CSPG4 (HV349014) at a concentration of 28 ug/ml overnight at 4°C. Afterward, the cells were incubated with a secondary antibody, Goat Anti-Rabbit IgG (Alexa Fluor-488) for 50 minutes at room temperature. The localization of CSPG4 was visualized (shown in green), and nuclear DNA was stained with DAPI (shown in blue).
