| Host species | Human |
| Isotype | IgG1-kappa |
| Clone ID | CAN10 |
| Biological activity | CAN10 effectively impairs the proinflammatory and profibrotic effects of IL1RAP-related cytokines (IL-1, IL-33, and IL-36) in human umbilical cord endothelial cells and human dermal fibroblasts, which are critical to systemic sclerosis pathophysiology [1]. CAN10 (mCAN10, mouse surrogate of CAN10) (10-20 mg/kg; i.p.; twice weekly; 7 weeks) reduces cutaneous and pulmonary fibrosis in a murine cGvHD model of systemic sclerosis, with significant improvements in dermal thickness, hydroxyproline content, myofibroblast counts, and lung Ashcroft scores [1]. |

| Background | Interleukin-1 receptor accessory protein (IL1RAP) is a ~65 kDa protein. Coreceptor for IL1RL2 in the IL-36 signaling system. Coreceptor with IL1R1 in the IL-1 signaling system. Associates with IL1R1 bound to IL1B to form the high affinity interleukin-1 receptor complex which mediates interleukin-1-dependent activation of NF-kappa-B and other pathways. Signaling involves the recruitment of adapter molecules such as TOLLIP, MYD88, and IRAK1 or IRAK2 via the respective TIR domains of the receptor/coreceptor subunits. Recruits TOLLIP to the signaling complex. |
