Biological activity Anti-LGALS9 (HFB200901) (10 ug/mL; 1 h) potently inhibits rLGALS9-induced vacuolization in MDCK and DLD-1 KRAS G12C mutant colon carcinoma cells [1]. HFB200901 (20 ug-10 mg; s.c.; once weekly; 8 weeks) reduces pancreatic LGALS9 expression, lowers peripheral and infiltrating Treg levels, increases pancreatic-infiltrating CD4 + T cells, and limits PanIN progression in KC mice [2].

Background

Galectin-9 (Gal-9) is a ~39 kDa protein. Binds galactosides. Has high affinity for the Forssman pentasaccharide. Ligand for HAVCR2/TIM3. Binding to HAVCR2 induces T-helper type 1 lymphocyte (Th1) death. Also stimulates bactericidal activity in infected macrophages by causing macrophage activation and IL1B secretion which restricts intracellular bacterial growth.
