ESAT-6 (EsxA) — Early Secretory Antigenic Target-6 is a 6 kDa protein secreted via the ESX-1 (Type VII) secretion system and is a primary virulence factor of M. tuberculosis. ESAT-6 forms a heterodimer with CFP-10 and is a key antigen in interferon-gamma release assays (IGRAs) such as QuantiFERON-TB Gold. Anti-ESAT-6 antibodies and recombinant proteins support IGRA development, T cell stimulation assays, and vaccine candidate evaluation.

CFP-10 (EsxB) — Culture Filtrate Protein-10 is the binding partner of ESAT-6 in the ESX-1 secretion complex. The ESAT-6/CFP-10 heterodimer is central to Mtb pathogenesis and serves as a differential diagnostic antigen (absent in BCG vaccine strains and most non-tuberculous mycobacteria). Ag85 Complex (Ag85A/B/C) — The Antigen 85 complex comprises three mycolyl transferases (Ag85A, Ag85B, Ag85C) essential for mycobacterial cell wall biosynthesis. Ag85B is among the most abundant secreted proteins of Mtb and a leading tuberculosis vaccine candidate antigen (e.g., H4:IC31, M72/AS01E).
