Background

Integrin alpha-4 (CD49d/ITGA4) is a ~114 kDa protein. Integrins alpha-4/beta-1 (VLA-4) and alpha-4/beta-7 are receptors for fibronectin. They recognize one or more domains within the alternatively spliced CS-1 and CS-5 regions of fibronectin. They are also receptors for VCAM1. Integrin alpha-4/beta-1 recognizes the sequence Q-I-D-S in VCAM1. Integrin alpha-4/beta-7 is also a receptor for MADCAM1.

    Immunofluorescence

Jurkat cells were fixed with 4% paraformaldehyde for 20 minutes, followed by blocking with 5% goat serum for 1 hour. The cells were then incubated with a primary antibody targeting Integrin alpha 4/CD49d (HY596043) at a concentration of 20 ug/ml overnight at 4°C. Afterward, the cells were incubated with a secondary antibody, Goat Anti-Rabbit IgG (Alexa Fluor-488) at a concentration of 6.5 ug/ml for 50 minutes at room temperature. The localization of Integrin alpha 4/CD49d was visualized (shown in green), and nuclear DNA was stained with DAPI (shown in blue).

    Flow cytometry

Flow-cytometry using anti-Human CD49d antibody. Jurkat cells were stained with an irrelevant antibody (Blue Histogram) or an anti-Human CD49d monoclonal antibody (Catalog HY596043, Yellow Histogram) at a concentration of 5 µg/ml for 30 mins at RT. After washing, bound antibody was detected using a Goat Anti-Rabbit IgG H&L Polyclonal Antibody, FITC (abinScience:TF690414) and cells analysed on a NovoCyte Flow Cytometer.
