| Background | Platelet endothelial cell adhesion molecule (CD31/PECAM1) is a ~82 kDa protein. Cell adhesion molecule which is required for leukocyte transendothelial migration (TEM) under most inflammatory conditions. Tyr-690 plays a critical role in TEM and is required for efficient trafficking of PECAM1 to and from the lateral border recycling compartment (LBRC) and is also essential for the LBRC membrane to be targeted around migrating leukocytes. Trans-homophilic interaction may play a role in endothelial cell-cell adhesion via cell junctions. Heterophilic interaction with CD177 plays a role in transendothelial migration of neutrophils. Homophilic ligation of PECAM1 prevents macrophage-mediated phagocytosis of neighboring viable leukocytes by transmitting a detachment signal. 1. Sachs, UJ. et al. Flow-cytometry using anti-human CD31 antibody. Jurkat cells were stained with an irrelevant antibody (Blue Histogram) or an anti-human CD31 monoclonal antibody (Catalog HB004023, Yellow Histogram) at a concentration of 5 µg/ml for 30 mins at RT. After washing, bound antibody was detected using a Goat Anti-Mouse IgG H&L Polyclonal Antibody, FITC (abinScience: MF690414) and cells analysed on a NovoCyte Flow Cytometer.

THP-1 cells were stained with an irrelevant antibody (Green Histogram) or an anti-human CD31 antibody monoclonal antibody (Catalog # HB004023 , Blue Histogram) at a concentration of 5 µg/ml for 30 mins at RT.
