Lassa virus (LASV) is an enveloped, bi-segmented, ambisense RNA arenavirus (~10.6 kb genome) and the causative agent of Lassa fever, a viral hemorrhagic fever endemic to West Africa with an estimated 100,000–300,000 infections and ~5,000 deaths annually. The multimammate rat (Mastomys natalensis) is the natural reservoir. LASV encodes four proteins on two genome segments: the glycoprotein precursor complex (GPC, cleaved into GP1 and GP2, sole surface antigen and vaccine/therapeutic target), nucleoprotein (NP, most abundant protein, diagnostic antigen and exonuclease/IFN antagonist), RNA-dependent RNA polymerase (L/RdRp), and the RING-domain matrix protein Z (drives budding). GPC is the exclusive target for neutralizing antibodies and is the immunogen in all LASV vaccine candidates currently in clinical development.

Glycoprotein Complex (GPC / GP1 / GP2) — Primary Neutralization Target
GPC is the sole surface antigen on LASV virions. It is synthesized as a precursor that is cleaved by SKI-1/S1P protease into GP1 (receptor binding via α-dystroglycan and LAMP1) and GP2 (class I fusion peptide). Z Protein (Matrix / RING Domain)
Z is a small RING-domain protein that drives LASV budding by recruiting the ESCRT machinery, analogous to VP40 in filoviruses. Z is also an IFN antagonist and a potential antiviral drug target (budding inhibitors). RdRp / L Protein (RNA Polymerase)
L protein is the large RNA-dependent RNA polymerase responsible for genome replication and transcription, and a target for broad-spectrum arenavirus antivirals (favipiravir/T-705 inhibits L activity). Nucleoprotein (NP)
NP encapsidates the viral RNA genome and possesses a unique 3'-5' exonuclease domain that degrades immunostimulatory dsRNA to suppress innate immunity. Why do you have so many anti-GPC antibody clones?
LASV GPC is a challenging target — natural LASV infection induces relatively weak neutralizing antibody responses compared to other hemorrhagic fever viruses. Having a panel of 17 clones targeting different GPC epitopes enables researchers to identify synergistic antibody combinations for therapeutic cocktail development, map neutralization-sensitive epitopes, and evaluate cross-lineage breadth (LASV has four major lineages with significant GP1 sequence divergence).
Can I work with these reagents at BSL-2?
Yes. All abinScience LASV products are recombinant proteins and antibodies — no live or inactivated virus material. Standard BSL-1/BSL-2 laboratory conditions are sufficient. Live LASV requires BSL-4 containment, but our recombinant reagents bypass this requirement entirely.
